How the reconstitution arithmetic works

Reconstitution in a laboratory setting is a dilution calculation. The lyophilized material in the vial has a nominal mass in milligrams, printed on the label and reported on the lot-matched Certificate of Analysis. When a measured volume of sterile diluent is added, the resulting concentration is simply mass divided by volume.

For example, a 10 mg vial reconstituted with 2 ml of sterile reconstitution solution gives 5 mg/ml, equivalent to 5000 mcg/ml. A 0.2 ml aliquot drawn from that prepared solution contains 1 mg of material. Halving the diluent volume doubles the concentration; doubling it halves the concentration.

  • Use the nominal mass on the vial label and confirm it against the Certificate of Analysis for that lot before calculating.
  • Measure the diluent volume with a calibrated device; the accuracy of the concentration figure is limited by the accuracy of the volume measurement.
  • Record the calculated concentration, the lot, and the preparation date in the laboratory record so later aliquots can be traced back to the certificate.
  • Follow the storage and handling protocol validated by the laboratory performing the work; lyophilized material and prepared solutions have different handling requirements.
